Determination of Soybean Oil, Protein and Amino Acid Residues in Soybean Seeds by High Resolution Nuclear Magnetic Resonance (NMRS) and Near Infrared (NIRS) Nature Precedings
I. C. Baianu; Tiefeng You; Doina Costescu; P R. Lozano; Valentin Prisecaru; Randall L. Nelson.
A detailed account is presented of our high resolution nuclear magnetic resonance (HR-NMR) and near infrared (NIR) calibration models, methodologies and validation procedures, together with a large number of composition analyses for soybean seeds. NIR calibrations were developed based on both HR-NMR and analytical chemistry reference data for oil and twelve amino acid residues in mature soybeans and soybean embryos. This is our first report of HR-NMR determinations of amino acid profiles of proteins from whole soybean seeds, without protein extraction from the seed. It was found that the best results for both oil and protein calibrations were obtained with a Partial Least Squares Regression (PLS-1) analysis of our extensive NIR spectral data, acquired...

Determinative atom model and the nature of chemical bond Nature Precedings
Wanchung Hu.
Quantum mechanics including Schrodinger and Dirac’s equations are used to describe the atom model. However, the basic assumption assuming electron energy can be described by E=h’w is wrong. If E=h’w, then the total energy of the electron will be E=rmv^2 in contrary to E=rmc^2 from special relativity. Here, I propose a new atom model integrating frame-dragging force, centrifugal force, electrostatic force, and magnetic force. In this new model, the proton group and neutron group in atomic nucleus are rotating in different direction. Thus, they provide equal and opposite frame-dragging force for orbiting electrons. The electrons are orbiting in the equator plane of nucleus. There is no net force or net acceleration for...

Determing Higher Fatty Acid Levels in Plant Materials NWISRL
Mayland, H.F..
The higher fatty acids (HFA) are important plant constituents that are implicated in the grass tetany hazard in livestock. A method is given whereby a technician can analyze 12 forage samples daily for HFA content. The method consists of saponifying the plant material in ethanol and KOH and extracting the subsequently acidified HFA with petroleum ether. The petroleum ether phase is evaporated and the HFA residue is dissolved in ethanol and then titrated with standardized isobutanolic KOH in the absence of Oxygen by using an Nitrogen atmosphere. A standard plant sample, analyzed over a 37-day period, had a mean of 136 mmol H+kg ± 4.5, where 0.1 mmol H+ℓ palmitic standard was determined with an accuracy of 99 ± 1.9%.

Did nature also choose arsenic? Nature Precedings
Felisa Wolfe-Simon; Paul C. W. Davies; Ariel Anbar.
All known life requires phosphorus (P) in the form of inorganic phosphate (PO4- or Pi) and phosphate-containing organic molecules. Pi serves as the backbone of the nucleic acids that constitute genetic material and as the major repository of chemical energy for metabolism in polyphosphate bonds. Arsenic (As) lies directly below P on the periodic table and so the two elements share many chemical properties, although their chemistries are sufficiently dissimilar that As cannot directly replace P in modern biochemistry. Arsenic is toxic precisely because As and P are similar...

Dielectric Constant of Liquids (Fluids) Shown to be Simple Fundamental Relation of Density over Extreme Ranges from - 50^o^ to + 600^o^C, Believed Universal Nature Precedings
William L. Marshall.
Examinations of past complex theoretical equations describing isothermal dielectric constants (_[epsilon]_) of liquids show that none describe experiments throughout and that only a two-constant equation of density is required. The dielectric susceptibility (_[epsilon]_ – 1) as a simple exponential function of density is shown here to describe _[epsilon]_ for both polar and non-polar organic liquids over all available experimental ranges from 0.50 to 1.2 g cm^-3^ and temperatures to 400^o^C. For water above 0.25 g cm^-3^, the equation describes the excellent dielectric-constant formulation of Fernández, et al., (1997) throughout its experimental range [-35 to +600^o^C; to 1000 MPa (10000 Atm)] and even when diluted by...

Dielectric Constant of Water Discovered to be Simple Function of Density over Extreme Ranges from – 35 to + 600^o^C and to 1200 MPa (12000 Atm.), Believed Universal Nature Precedings
William L. Marshall.
The experimental static dielectric constants (_[epsilon]_) of water formulated by Fernández, et al., (1997-International Association for the Properties of Water and Steam, IAPWS), –35 to 600^o^C; 0.1 to 1200 MPa, are discovered to be closely described throughout by a simple equation of density (_[rho]_), presented here, and agreeing with their reported calculated (extrapolated) values to 923^o^C. Eight limiting boundary conditions are observed. Values of _[epsilon]_ follow isothermal straight-line slopes of log (_[epsilon]_ – 1) vs log _[rho]_ down to densities of 0.25 g cm^-3^ water, even in dioxane-water solutions. At lower densities, all slopes curve asymptotically in approaching unity, while becoming unity throughout...

Discovery of multiple lead compounds as M2 inhibitors through the focused screening of a small primary amine library. Nature Precedings
Wenhui Hu; Shaogao Zeng; Chufang Li; Yanling Jie; Zhiyuan Li; Ling Chen.
The discovery of new anti-influenza drugs is urgent, particularly considering the recent threat of swine flu. In this study, the influenza virus M2 protein was expressed in HEK293 cells and shown to have selective ion channel activity for monovalent ions. The anti-influenza virus drug amantadine hydrochloride significantly attenuated the inward current induced by hyperpolarization of HEK293 cell membranes. Although adamantine derivatives are the only M2 drugs for influenza virus A, their use is limited in the US due to drug resistance. Here we report the discovery of multiple M2 inhibitor lead compounds that were rapidly generated through focused screening of a small primary amine library. The screen was designed using a scaffold-hopping strategy based on...

Disruptive Strategies for Removing Drug Discovery Bottlenecks Nature Precedings
Sean Ekins; Chris L. Waller; Mary P. Bradley; Antony J. Williams.
Drug Discovery is shifting focus from the industry to outside partners and in the process creating new bottlenecks, suggesting the need for a more disruptive overhaul. Technologies like high throughput screening (HTS) have moved to a larger number of academic and institutional laboratories in the US, with little apparent coordination or consideration of the outputs and creating a translational gap. While there have been collaborative public private partnerships in Europe to share pharmaceutical data, the USA has lagged behind. Sharing precompetitive computational models may be the next frontier to provide more confidence in the quality of the leads produced and attract investment. We suggest there needs to be an awareness of what research is going on in...

Docking and QSAR Studies of Camptothecin Derivatives as Inhibitor of DNA Topoisomerase-I Nature Precedings
Dharmendra K. Yadav; Feroz Khan; Santosh K. Srivastava.
Camptothecin (CPT) is a cytotoxic quinoline alkaloid which inhibits the DNA enzyme Topoisomerase-I (Topo-I) and has shown remarkable anticancer activity in preliminary clinical trials. The major limitation is its low solubility and high adverse reaction. In the studied work, we performed molecular docking of CPT derivatives against Topo-I and developed the quantitative structure activity relationship (QSAR) model for anticancer activity screening. For QSAR, we used CPT and other anticancer drugs with its IC50 values. We used a total of forty seven anticancer drugs as training set and eight compounds as test set and thirty derivatives of CPT as query set. Total of fifty two chemical descriptors were used for the quantitative data calculation. Only four...

Dust in the wind – drift of dust containing insecticides – a risk for honey bees (_Apis mellifera_ L.)? Nature Precedings
Pablo T. Georgiadis; Jens Pistorius; Udo Heimbach.
In late April 2008, dust drift containing insecticide resulted in the largest bee poisoning in Germany for 30 years. The reason for these incidents was the contamination of flowering bee forage plants with dust particles abraded from maize seeds treated with Clothianidin recorded in the Upper Rhine Valley, Baden-Wuerttemberg and in parts of Bavaria during sowing of maize.

Clothianidin, an insecticidal active substance from the group of neonicotinoids, works as agonist at the nicotinic acetylcholine receptors with broad spectrum of efficacy. Clothianidin is also used as a seed dressing of maize (agent for seed treatment “Poncho Pro®”; 1,.25 mg a.i. Clothianidin./kernel) for the control of the...
